Provinase® is a powerful, non-specific endonuclease enzyme that cleaves both DNA and RNA—regardless of sequence—into small oligonucleotides. Its mechanism is designed to efficiently degrade contaminating nucleic acids in a wide range of biological and bioprocessing applications.

Mechanism of Action:

  1. Nucleic Acid Binding
    Provinase® identifies and binds to exposed double-stranded or single-stranded DNA and RNA present in a solution, such as those released from host cells during bioproduction or sample preparation.
  2. Phosphodiester Bond Hydrolysis
    It then catalyzes the hydrolysis of phosphodiester bonds within the nucleic acid backbone, fragmenting long DNA/RNA strands into short oligonucleotides or mononucleotides.
  3. Rapid and Non-Sequence-Specific Cleavage
    Unlike restriction enzymes, Provinase® does not require a specific recognition sequence, making it effective across diverse sample types and species. It acts quickly and uniformly across nucleic acid types.
  4. End Product
    The degraded nucleic acids are rendered biologically inactive and are easily removed via filtration, chromatography, or centrifugation, contributing to lower viscosity, higher purity, and more efficient downstream processing.

What is Provinase®?

Provinase® is a genetically engineered non-specific nuclease derived from Serratia marcescens and expressed in E. coli under cGMP conditions. It exhibits high activity and broad specificity, effectively degrading all forms of nucleic acids—including single- and double-stranded, linear, and circular structures.

How does Provinase® work?

Provinase® is a homodimeric enzyme composed of two 30 kDa subunits stabilized by essential disulfide bonds, ensuring its activity and structural integrity. It hydrolyzes internal phosphodiester bonds in nucleic acids, generating 5′-monophosphate oligonucleotides typically 2–5 bases long.

 

What is Provinase® good for?

Biopharmaceutical products such as lentiviral and AAV vectors, antibody therapeutics, vaccines, and recombinant proteins are produced using continuously passaged cell lines. Despite rigorous purification, residual host cell nucleic acids may persist, posing risks such as immunogenicity, pathogenicity, or tumorigenesis. Therefore, it is essential to reduce these nucleic acids to safe, regulatory-compliant levels in the final drug product.

What is the Enzyme activity of Provinase®?

One unit of activity corresponds to the amount of enzyme required to change the absorbance at 260 nm by 1.0 under the condition of excess herring sperm DNA at 37°C and pH 8.0 for 30 minutes (equivalent to the amount of enzyme required to completely digest 37µg of DNA substrate). The verified enzyme activity of Provinase® is ≥ 250 U/µL.
